Thermoplastic polyurethane (TPU) has been widely applied in various medicaldevices, such as artificial blood vessels, dialysis pumps and catheters, due to itsgood mechanical and biocompatible properties. However, bacterial infectionoccurs often in urinary and central venous catheters, which is attributed to thehydrophobic property of TPU, leading to non-specific adsorption of bacteria.Meanwhile, placement of catheters can be uncomfortable and painful for apatient. It has been found that zwitterionic phosphatidylcholine materials possessexcellent biocompatible, lubricant and antifouling properties. In this proposal, weattempt to develop an efficient and facile way to graft zwitterionic antifouling andhydrophilic medical coatings on TPU by dip coating and photoreaction in anambient environment. Amphiphilic copolymers, poly(2-methacryloyloxyethylphosphorylcholine), PMPC, were covalently grafted onto TPU substrate throughphotografting of the ketone structure in photoinitiator under UV irradiation. We willadopt two modification methods, semi-interpenetrating polymer network andgraft-onto approach, to ensure the success and most-cost effeciency of theproject. The chemical structures of polymers and surface coatings will becharacterized by nuclear magnetic resonance (NMR), Gel permeationchromatography (GPC), attenuated total reflection-Fourier-transform infraredspectroscopy (ATR-FTIR), X-ray photoelectron spectroscopy (XPS), contactangle goniometer, ellipsometry, and atomic force microscopy (AFM). Functions ofPMPC-modified TPU tubes will be examined by the friction test, anti-proteinadhesion, and antibacterial adhesion in comparison with un-modified tubes.Furthermore, tests for biocompatibility will be performed and certificated forcommercialization.
